BACKGROUND: Receptor-interacting serine-threonine kinase 3 (RIPK3)-mediated necroptosis has been implicated in the progression of myocardial infarction (MI), but the underlying mechanisms, particularly whether microRNAs (miRNAs) are involved, remain largely unknown. RESULTS: A microarray analysis was used to screen for miR-325-3p expression in myocardial tissues from MI mice, and the expression was confirmed with qRT-PCR. The levels of myocardial enzymes were measured using commercial kits, and an echocardiography system was utilized for the detection of cardiac function parameters. The pathological features and infarction sizes of cardiac tissues were examined using H&E, TCC and Masson's trichrome staining, and the amount of cell apoptosis was determined using an in situ TUNEL assay. Cardiomyocytes were isolated and then subjected to hypoxia induction in vitro. The expression of the RIPK1, RIPK3 and phosphorylated MLKL (p-MLKL) proteins was measured using a Western blot. The mouse cardiomyocyte cell viability was analyzed by an MTT assay. The mRNA target of miR-325-3p was predicted using TargetScan v7.2 and then validated using a dual-luciferase reporter assay. The overexpression of miR-325-3p evidently decreased the expression levels of lactate dehydrogenase (LDH), phosphocreatine kinase (CK), superoxide dismutase (SOD) and malondialdehyde (MDA), inhibited left ventricular end-diastolic diameter (LVEDD) and left ventricular end-systolic diameter (LVESD), and promoted left ventricular ejection fraction (LVEF) and left ventricular fractional shortening (LVES). In addition, miR-325-3p overexpression attenuated the degree of injury to the cardiac tissue, decreased the infarct sizes and downregulated the expression of the necrosis-related proteins RIPK1, RIPK3 and p-MLKL. CONCLUSIONS: The RIPK1/RIPK3/p-MLKL axis-induced necroptosis that occurred during MI was mediated by a miRNA module, miR-325-3p, which can effectively ameliorate the symptoms of MI by suppressing the expression of RIPK3.

OBJECTIVE: This study aims to compare the clinical effects of selective interventional therapy (PCI) under the guidance of fractional flow reserve (FFR) and coronary arteriography. METHODS: Patients with sub-acute ST-segment elevation myocardial infarction (sub-acute STEMI), who were under selective PCI treatment between April 2012 and June 2014, were included into this study. These patients were divided into two groups, based on FFR measurements: FFR-PCI group and radiography-PCI group. Then, differences in clinical symptoms, coronary angiography, intervention, and endpoint events were compared between these two groups. RESULTS: A total of 592 patients with sub-acute STEMI were included in this study (207 patients in the FFR-PCI group and 385 patients in the radiography-PCI group). No statistical differences were observed in baseline clinical data and coronary angiography results between these two groups. Mean stent number was greater in the radiography-PCI group (1.22 ± 0.32) than in the FFR-PCI group (1.10 ± 0.29), and the difference was statistically significant (P = 0.019). During the follow-up period, 78 adverse events occurred (21 adverse events in the FFR-PCI group and 57 adverse events in the radiography-PCI group); and no statistical significance was observed between these two groups (log-rank P = 0.112). CONCLUSION: Selective PCI treatment in patients with sub-acute STEMI under FFR acquired similar effects, compared to PCI treatment under the guidance of radiography, which can reduce the mean stent number.

BACKGROUND AND AIMS: Down-regulation of the growth arrest specific transcript 5 (GAS5) (long non-coding RNA) is associated with cell proliferation of gastric cancer (GC) and a poor prognosis. We aimed to investigate whether the variant rs145204276 of GAS5 is associated with the prognosis of GC in the Chinese population, and to unveil the regulatory mechanism underlying the GAS5 expression in GC tissues. METHOD: 1,253 GC patients and 1,354 healthy controls were included. The frequency of the genotype del/del and the allele del of rs145204276 were compared between the patients and the controls and between different subgroups of patients classified by clinicopathological variables. The overall survival rate was analyzed according to the Kaplan-Meier method using the log-rank test. RESULTS: The frequency of genotype del/del was significantly lower in patients than in the controls (7.0% vs. 9.1%, p = 0.001). Kaplan-Meier analysis showed that genotype del/del was significantly associated with a higher survival rate (p = 0.01). Patients with late tumor stage were found to have a significantly lower rate of genotype del/del than those with an early tumor stage (4.9% vs. 8.8%, p = 0.01). Patients with UICC III and IV were found to have a significantly lower rate of genotype del/del than those with UICC I and II (5.3% vs. 8.1%, p = 0.02). CONCLUSION: The variant rs145204276 of GAS5 is associated with the development and prognosis of GC. The allele del of rs145204276 is associated with a remarkably lower incidence of cancer progression and metastasis.

AIMS: A successful cephalosomatic anastomosis ("head transplant") requires, among others, the ability to control long-term immune rejection and avoidance of ischemic events during the head transference phase. We developed a bicephalic model of head transplantation to study these aspects. METHODS AND RESULTS: The thoracic aorta and superior vena cava of a donor rat were anastomosed with the carotid artery and extracorporeal veins of a recipient rat by vascular grafts. Before thoracotomy in the donor rat, the axillary artery and vein of the donor were connected to the carotid and the extracranial vein of the third rat through a silicone tube. The silicone tube was passed through a peristaltic pump to ensure donor brain tissue blood supply. There is no ischemia reperfusion injury in donor brain tissue analyzed by electroencephalogram. Postoperative donor has pain reflex and corneal reflex. CONCLUSIONS: Peristaltic pump application can guarantee the blood supply of donor brain tissue per unit time, while the application of temperature change device to the silicone tube can protect the brain tissue hypothermia, postoperative experimental data show that there is no brain tissue ischemia during the whole operation. The application of vascular grafting can also provide the possibility of long-term survival of the model.

BACKGROUND: Previous studies have investigated the associations between interleukin-10 (IL-10) gene polymorphisms (-592C/A, -819C/T and -1082G/A) and risk of coronary artery disease (CAD). However, the results were inconsistent. The aim of this study was to clarify the relationship between IL-10 polymorphisms and CAD risk by a meta-analysis approach. METHODS: The PubMed, Embase, Web of Science, China National Knowledge Infrastructure (CNKI) and Wanfang databases were searched according to predefined criteria for all relevant studies published before June 1, 2015. Pooled odds ratios (ORs) and 95% confidence intervals (95% CIs) were computed to assess the association. RESULTS: 24 eligible studies were enrolled including 9736 CAD patients and 8606 controls. We observed a significant decreased risk of CAD for IL-10 -819C/T polymorphism (T allele vs. C allele:OR = 0.91, 95% CI = 0.84-0.99; TT vs. CT + CC:OR = 0.82, 95% CI = 0.69-0.98), especially in Asians (T allele vs. C allele:OR = 0.76, 95% CI = 0.60-0.96; TT vs. CC:OR = 0.51, 95% CI = 0.27-0.96; TT vs. CT + CC:OR = 0.62, 95% CI = 0.44-0.88). Moreover, we found IL-10 -1082G/A polymorphism might contribute to an increased CAD risk in Asians (AA vs. GG:OR = 1.89, 95% CI = 1.36-2.64; AA vs. AG + GG:OR = 1.39, 95% CI = 1.14-1.68) but not in other ethnic groups. However, no significant association between the IL-10 -592C/A polymorphism and CAD risk was observed. CONCLUSIONS: Our results indicated that IL-10 -819C/T and IL-10 -1082G/A polymorphisms significantly and race-specifically correlate with CAD risk.

Vascular extracellular matrix (ECM) remodelling, which is the result of disruption in the balance of ECM synthesis and degradation, induces vessel fibrosis and thereby leads to hypertension. Leptin is known to promote tissue fibrosis, while adiponectin has recently been demonstrated to be anti-fibrogenic in tissue fibrosis. In this study, we aimed to evaluate the leptin-antagonist function of adiponectin and to further elucidate the mechanisms through which adiponectin dampens leptin signalling in vascular smooth muscle cells, thus preventing excess ECM production, in our already established 3D co-culture vessel models. Our 3D co-culture vessel model, which mimics true blood vessels, is composed of vascular endothelial cells, vascular smooth muscle cells and collagen type I. We validated the profibrogenic effects of leptin and analysed matrix metalloproteinase 2 (MMP2), MMP9, tissue inhibitor of metalloproteinase 1 (TIMP1) and collagen types II/IV secretion in 3D vessel models. The protective/inhibitory effects of adiponectin were re-analysed by inhibiting adiponectin receptor 1 (AdipoR) and AdipoR2 expression in endothelial cells using RNAi technology. In the 3D vessel models, adiponectin blocked the leptin-stimulated secretion of collagen types II/IV and TIMP1 while significantly increasing MMP2/9 activity. In endothelial cells, adiponectin induced phosphorylation of AMPK, thereby suppressing leptin-mediated STAT3 phosphorylation through induction of SOCS3 in smooth muscle cells. Our findings indicate that adiponectin disrupted the leptin-induced vascular ECM remodelling via AdipoR1 and enhanced AMPK signalling in endothelial cells, which, in turn, promoted SOCS3 up-regulation in smooth muscle cells to repress leptin-stimulated phosphorylation of STAT3.

An efficient inorganic polymer coagulant, poly-aluminum-ferric-silicate-chloride (PAFSC), was developed using two approaches: (i) hydroxylation of the mixture of AlCl3, FeCl3 and fresh polysilicic acid in different Al/Fe/Si molar ratios to obtain PAFSCc and (ii) hydroxylated poly-aluminum-iron-chloride (PAFC) combined with aged polysilicic acid in different Al/Fe/Si ratios to produce PAFSCm. The properties of PAFSC in comparison with polyaluminum silicate chloride (PASC) and polyferric silicate chloride (PAFC) were characterized by various experimental methods. The effect of Al/Fe/Si molar ratio on the hydrolysis-polymerization process of Al (III) and Fe (III) in PAFSC solutions was examined by pH titration, and the effect of Al/Fe/Si molar ratio on electrokinetic mobility of PAFSC was studied by Zeta potential measurement. The laboratory experiments were performed to evaluate the PAFSC in comparison with polyaluminum chloride (PAC) for the coagulation of synthetic water samples, actual surface water and wastewater. The results show that interactions exist among aluminum species, ferric species and polysilicic acid, and the Al/Fe/Si molar ratio affects the Zeta potential of the hydrolyzate and the coagulating performance. PAFSC achieved a better water treatment result than PAC. At the same basicity (B) value and Al/Fe/Si ratio, PAFSCc has better coagulation performance than PAFSCm. PAFSC is a new type and high efficiency composite inorganic polymer coagulant.

A polyaluminum chloride (PAC) coagulant was prepared from AlCl3 x 6H2O and Na2CO3. The Al13 species in PAC was separated and purified by the SO4(2-)/Ba2+ deposition-replacement method, and characterized by 27Al-NMR and XRD. From 27Al-NMR spectroscopy, it was found that PAC obtained after separation and purification contained more Al13 (PAC-Al13, for short) than original PAC before separation and purification. In XRD spectra, a strong Al13 signal appeared in the range of 2theta from 5 to 25 degrees. Jar tests were performed to test the coagulation efficiency of AlCl3, PAC and PAC-Al13 in treating synthetic or actual water samples. Compared with PAC and AlCl3, PAC-Al13 gives the best results for turbidity, humic acid and color removal, and achieves the highest charge-neutralizing ability. Under the study conditions, PAC-Al13 gave about 90% humic acid removal and almost 100% reactive blue dye removal when its dosages were 4.0 and 15 mg/L as Al, respectively. The Al13 species has a higher positive charge and is the most effective polymeric Al species in water and wastewater treatment.

A series of poly-aluminum-chloride-sulfates (PACS), which have different OH/Al (gamma) and Al(3+)/SO4(2-) mole ratios, has been prepared using AlCl3 x 6H2O, A(SO4)3 x 18H2O and Na2CO3 as raw materials. The electrophoretic nature of PACS was investigated by electrophoresis. Laboratory experiments were undertaken to evaluate the PACS in comparison with polyaluminum chloride (PAC) for the coagulation of simulating water and actual wastewaters. The experimental results show that the gamma value and the Al(3+)/SO4(2-) mole ratio affect the electrophoretic nature of PACS. PACS has a maximum zeta potential at about a gamma value of 1.5 and Al(3+)/SO4(2-) mole ratio of 12-16. The zeta potential of PACS varies with pH. The performance of PACS as coagulant is affected by gamma value and Al(3+)/SO4(2-) mole ratio. PACS of gamma = 2.1 and Al(3+)/SO4(2-) = 15 gives best turbidity removal effectiveness. In comparison with PAC of gamma = 2.0, PACS of gamma = 2.0 and Al(3+)/SO4(2-) = 16 gives higher removal efficiency for turbidity and COD, and shows the following advantages in the clarification of waters and wastewaters: rapid aggregation velocity, larger and heavier flocs, and lower required dosage.

The electrokinetic characteristics and coagulation behaviors of polyaluminum silicate chloride (PASiC) and polyaluminum chloride (PAC) were studied and compared by streaming current (SC) measurement and jar test method. The experimental results showed that the interaction between polysilicic acid characterized negative charge and hydrolyzed aluminum species result in a decrease of the charge-neutralizing ability of PASiC, compared to PAC. The decrease has a close relationship with the basicity (B) and Al/Si molar ratio in PASiC. The less the B value and the Al/Si molar ratio, the lower the charge-neutralizing ability of PASiC is. In contrast, the preparation technique for PASiC affects the charge - neutralization of PASiC to a smaller extent. In addition, compared with PAC, PASiC may enhance aggregating efficiency and give better coagulating effects.
